Gene Hlac_2868 in replicon pHL500

Hlac_2868 codes for a family 42 beta-galactosidase enzyme. It was first investigated using bioinformatics to compare it to homologous haloarchaeal proteins. See DasSarma et al. Plos One. 2013 Mar;8(3):e58587. The enzyme was then cloned, overexpressed, purified, and characterized using NRC-1 as a haloarchaeal expression system. See Karan et al. BMC Biotech. 2013 Jan;13(3). In order to determine the source of the enzyme’s cold adaptation the bioinformatic and genetic approaches were combined to perform site directed single amino acid mutations and it was determined that single amino acid changes can affect the cold adaptation of the enzyme.
